    The single-cell green alga <italic>Chlamydomonas reinhardtii</italic> possesses two α-tubulin genes (<italic>tua1</italic> and <italic>tua2</italic>) and two β-tubulin genes (<italic>tub1</italic> and <italic>tub2</italic>), with the two genes in each pair encoding identical amino acid sequences. Here, we screened an insertional library to establish eight disruptants with defective <italic>tua2</italic>, <italic>tub1</italic>, or <italic>tub2</italic> expression. Most of the disruptants did not exhibit major defects in cell growth, flagellar length, or flagellar regeneration after amputation. Because few tubulin mutants of <italic>C</italic>. <italic>reinhardtii</italic> have been reported to date, we then used our disruptants, together with a <italic>tua1</italic> disruptant obtained from the <italic>Chlamydomonas</italic> Library Project (CLiP), to isolate tubulin-mutants resistant to the anti-tubulin agents propyzamide (pronamide) or oryzalin. As a result of several trials, we obtained 8 strains bearing 7 different α-tubulin mutations and 12 strains bearing 7 different β-tubulin mutations. One of the mutations is at a residue similar to that of a mutation site known to confer drug resistance in human cancer cells. Some strains had the same amino acid substitutions as those reported previously in <italic>C</italic>. <italic>reinhardtii</italic>; however, the mutants with single tubulin genes showed slightly stronger drug-resistance than the previous mutants that express the mutated tubulin in addition to the wild-type tubulin. Such increased drug-resistance may have facilitated sensitive detection of tubulin mutation. Single-tubulin-gene disruptants are thus an efficient background of generating tubulin mutants for the study of the structure–function relationship of tubulin.

    Novel actin-like protein (NAP) is a highly divergent actin expressed in Chlamydomonas. With its low sequence similarity, it is uncertain whether NAP can polymerize into filaments. Here I assessed it by ectopically expressing enhanced green fluorescent protein-tagged NAP (EGFP-NAP) in cultured cells. EGFP-NAP was excluded from stress fibres but partially co-localized with endogenous actin in the cell periphery. In fluorescence recovery after photobleaching experiment, turnover rate of EGFP-NAP was similar to the estimated diffusion rate of monomeric actin. Therefore, EGFP-NAP likely accumulates by diffusion. These findings suggest that NAP has extremely poor ability to polymerize. (C) 2011 Elsevier Inc. All rights reserved.

    Actin is an ancient cytoskeletal protein that plays many essential roles in cell motility. In eukaryotes, its gene belongs to a highly conserved gene family, while the protein is also a member of an actin superfamily comprising various kinds of actin-related proteins (Arps). A ciliate, Tetrahymena, has a unique conventional actin. Data from the TIGR Tetrahymena genome project and our own research suggest the existence of 12 actin-like sequences: four conventional actins, two of Arp4, one each of Arp1, Arp2, Arp3, Arp5, and Arp6, and a novel actin-related protein, tArp. We cloned the entire cDNA sequences of Tetrahymena Arp2 (tArp2), Tetrahymena Arp3 (tArp3), and tArp for the work described herein. In phylogenetic analyses, tArp was not included in any Arp, subfamily. Unlike other known Arps, tArp localizes in cilia, and its expression was upregulated after deciliation. To see the precise localization of tArp, cilia were fractionated and analyzed using specific antibodies. tArp was observed preferentially in the "outer-doublet" fraction, while actin was found in the "crude-dynein" fraction. In immunoelectron microscopy, most of the gold particles were found either on the outer-doublet or central-pair microtubules. These results suggest that tArp is a ciliary component and that it has a unique function in the formation and maintenance of cilia.

    Chlamydomonas has two actin genes: one encoding a conventional actin (90% amino acid identity with mammalian actin) and the other a highly divergent actin (NAP; 64% identity). The expression of the two genes is regulated in a mutually exclusive manner. Thus, ida5, a mutant that lacks the conventional actin (CrA) gene, expresses NAP abundantly, whereas wild-type cells express NAP only negligibly under normal conditions. To explore the physiological significance of the two actins, chimeric genes with the 5' upstream region of one gene replaced by that of the other were constructed and used to transform ida5. The transformant (TF5) with a chimeric clone comprising the 5'-untranslated region from the NAP gene and the CrA-encoding sequence recovered the dyneins missing in ida5 and showed almost normal motility. After deflagellation of this transformant, however, only about 30% of cells grew flagella, unlike wildtype cells, &gt; 80% of which displayed reflagellation. Transformant TF10, which contains the CrA upstream region and NAP coding region, underwent reflagellation normally, as did the parent strain, ida5. In TF5, the mRNA level of both CrA and NAP was increased greatly during reflagellation. In light of the recent finding that NAP mRNA is expressed transiently upon reflagellation in wild-type cells, the described results suggest that 1) the expression of NAP mRNA is indispensable for flagellation and 2) robust expression of CrA may inhibit proper flagellation by interfering with the function of NAP in the early stages of reflagellation.

    The unicellular green alga Chlamydomonas reinhardtii has two actin genes, one encoding a conventional actin (90% amino acid identity with mammalian actin), the other a highly divergent actin (64% identity) named novel actin-like protein (NAP). To see whether the presence of conventional and unconventional actins in a single organism is unique to C. reinhardtii, we searched for genomic sequences related to the NAP sequence in several other species of volvocalean algae. Here we show that Chlamydomonas moewusii and Volvox carteri also have, in addition to a conventional actin, an unconventional actin similar to the C. reinhardtii NAP. Analyses of the deduced protein sequences indicated that the NAP homologues form a distinct group derived from conventional actin.

    Chlamydomonas has two actin genes, one coding for a conventional actin and the other coding for a highly divergent actin. The divergent actin NAP (for "novel actin-like protein") is expressed only negligibly in wild-type cells but abundantly in a null mutant of conventional actin, the ida5 mutant. The presence of the dormant NAP gene suggests that NAP may also have its own function in wild-type cells under some conditions. However, no specific functions have been suggested. In this study, we examined the expression of actin and NAP in wild-type and ida5 cells under conditions where actin function has been shown to be important. We found that deflagellation induces the expression of NAP as well as that of actin in wild-type cells. The expressed NAP becomes localized to the regrown flagella, apparently without being associated with dynein. Mating of gametes also increased the expression of actin in wild-type cells and that of NAP in ida5 cells, resulting in accumulation of these proteins in flagella (in both wild-type and ida5 cells) and the fertilization tubule (only in wild-type cells). However, it did not induce significant NAP expression in wild-type cells. These and other observations suggest that the expression of actin and NAP mRNAs is controlled by two discrete mechanisms and that NAP plays a role in flagellar formation in wild-type cells.

    The ida5 mutant of Chlamydomonas, first isolated as a mutant lacking a subset of axonemal inner-arm dyneins, has recently been shown to lack conventional actin owing to a serious mutation in its gene. It lacks inner-arm dyneins probably because actin is an essential subunit for their assembly. In addition, male gametes of ida5 are unable to produce the fertilization tubule, a structure that contains a core of actin filament bundles. To establish that those observed deficiencies are solely attributable to the loss of actin, and to provide a basis for future studies on the actin function in this organism, we examined in this study whether transformation of this mutant with cloned actin genes can rescue the mutant phenotypes. Cotransformation of the double mutant ida5arg2 with the wild-type actin gene and arginino-succinate lyase gene that suppresses the arg2 mutation yielded several transformants that displayed increased motility. All of them were found to have acquired the introduced actin gene In the genome and the product actin in the flagella, and regained the missing inner-arm dyneins and wild-type motility. In addition, most transformants also became able to grow the fertilization tubule when mating reaction was induced. In addition to the wild-type actin gene, we also used a chimeric actin gene in which the N-terminal Y2 amino-acid sequence of Chlamydomonas actin was replaced by that of the greatly divergent Tetrahymena actin. Transformants with this gene also resulted in recovery of inner-arm dynein and 70-80% of the wild-type level of motility. These results established that the lack of inner-arm dynein and the fertilization tubule in ida5 are consequences of its loss of conventional actin. Furthermore, they demonstrate that Chlamydomonas offers an excellent experimental system with which to study the structure-function relationship of actin by means of mutant analysis.

    The Chlamydomonas mutant ida5 is deficient in the conventional actin gene and its axoneme lacks a subset of inner dynein arms that contain actin as a subunit. However, this mutant retains some other inner dynein arms because a novel protein (NAP) is expressed as a substitute for actin. In this study, we show by sequence analysis that NAP is identical to a putative actin-related protein, the cDNA sequence of which has recently been reported and shown to have 64% amino acid identity with conventional actin. A polyclonal antibody raised against a synthetic polypeptide corresponding to the NH2-terminal sequence of this protein specifically reacted with the spot corresponding to NAP in two-dimensional electrophoresis patterns. NAP apparently can substitute for conventional actin in some, but not all, cellular functions, and therefore can be regarded as a highly divergent actin. This unconventional actin appears to be expressed only when conventional actin is absent. (C) 1998 Academic Press.

    Chlamydomonas flagellar inner-arm dynein consists of seven subspecies (a-g), of which all but f contain actin as subunits. The mutant ida5 and a new strain, ida5-t, lack four subspecies (a, c, d, and e). These mutants were found to have mutations in the conventional actin gene, such that its product is totally lost; ida5 has a single-base deletion that results in a stop codon at a position about two-thirds from the 5' end of the coding region, and idn5-t lacks a large portion of the entire actin gene. Two-dimensional gel electrophoresis patterns of the axonemes and inner-arm subspecies b and g of ida5 lacked the spot of actin (isoelectric point [pI] = similar to 5.3) but had two novel spots with pIs of similar to 5.6 and similar to 5.7 instead. Western blot with different kinds of anti-actin antibodies suggested that the proteins responsible for the two novel spots and conventional actin are different but share some antigenicity. Since Chlamydomonas has been shown to have only a single copy of the conventional actin gene, it is likely that the novel spots in ida5 and ida5-t originated from another gene(s) that codes for a novel actin-like protein(s) (NAP), which has hitherto been undetected in wildtype cells. These mutants retain the two inner-arm subspecies b and g, in addition to f, possibly because NAP can functionally substitute for the actin in these subspecies while they cannot in other subspecies. The net growth rate of ida5 and ida5-t cells did not differ from that of wild type, but the mating efficiency was greatly reduced. This defect was apparently caused by deficient growth of the fertilization tubule. These results suggest that NAP can carry out some, but not all, functions performed by conventional actin in the cytoplasm and raise the possibility that Chlamydomonas can live without ordinary actin.

    Using the CHO2 monoclonal antibody raised against CHO spindles (Sellitto, C., M. Kimble, and R. Kuriyama. 1992. Cell Motil. Cytoskeleton. 22:7-24) we identified a 66-kD protein located at the interphase centrosome and mitotic spindle. Isolated cDNAs for the antigen encode a 622-amino acid polypeptide. Sequence analysis revealed the presence of 340-amino acid residues in the COOH terminus, which is homologous to the motor domain conserved among other members of the kinesin superfamily. The protein is composed of a central alpha-helical portion with globular domains at both NH2 and COOH termini, and the epitope to the monoclonal antibody resides in the central alpha-helical stalk. A series of deletion constructs were created for in vitro analysis of microtubule interactions. While the microtubule binding and bundling activities require both the presence of the COOH terminus and the alpha-helical domain, the NH2-terminal half of the antigen lacked the ability to interact with microtubules. The full-length as well as deleted proteins consisting of the COOH-terminal motor and the central alpha-helical stalk supported microtubule gliding, with velocity ranging from 1.0 to 8.4 mu m/minute. The speed of microtubule movement decreased with decreasing lengths of the central stalk attached to the COOH-terminal motor. The microtubules moved with their plus end leading, indicating that the antigen is a minus end-directed motor. The CHO2 sequence shows 86% identify to HSET, a gene located at the centromeric end of the human MHC region in chromosome 6 (Ando, A., Y. Y. Kikuti, H. Kawata, N. Okamoto, T. Imai, T. Eki, K. Yokoyama, E. Soeda, T. Ikemura, K. Abe, and H. Inoko. 1994. Immunogenetics. 39:194-200), indicating that HSET might represent a human homologue of the CHO2 antigen.

    Two novel Chlamydomonas mutants, ida5 and ida6, that lack subsets of inner-arm dynein have been isolated and mapped to discrete loci on the right arm of linkage group XIV. Of the seven different inner-arm dynein subspecies (a, b, c, d, e, f and g) identified by ion-exchange chromatography, ida5 lacks a, c, d and e, while ida6 lacks e alone; these are the only mutants that have been shown to lack subspecies e. Both strains can swim, albeit more slowly than the wild type. Hence, subspecies e must contribute-to flagellar movement although it is unnecessary for the generation of undulating movement.

    Microtubules are assembled from tubulin dimers that are arranged in a semi-crystal lattice with high regularity. Dynamics of tubulin dimer within microtubules is thus expected to be directly affecting the stability of microtubules, however, it has been difficult to quantitatively describe such properties. By analyzing the time course of pattern changes in X-ray fiber diffractions from aligned microtubules with a high-flux synchrotron beam of BL40XU (SPring-8), we found microtubules showed rapid elongation in the axial tubulin repeat and the concomitant increase of structural flexibility with a time constant of about 0.2 s after applying paclitaxel, microtubule-stabilizer. Contrasting effects were found for laulimalide, another type of microtubules stabilizer.

    クラミドモナスには保守的配列のアクチンと、極めてdivergentな配列のNAPの2種のアクチン遺伝子が存在し、互いに異なる発現制御を受けている。両者の機能的差異と相関を理解することが重要と考えられるが、NAP独自の機能やその起源は謎である。今年度は昨年に引き続き、キメラ遺伝子による両者の人為的発現制御の効果を検討するとともに、オオヒゲマワリ目におけるNAPホモログを同定し、分子系統的解析を行った。
    1.キメラ遺伝子の導入によるNAP・アクチンの人為的発現制御
    翻訳開始点(ATG)を境に、NAPの上流領域(発現制御領域)とアクチンのタンパク質コード領域をつなげたキメラ遺伝子(pNAPΔpA7)、およびアクチンの上流領域とNAPのコード領域をつなげた遺伝子(pA7ΔpNAP)をそれぞれ作成し、アクチン欠損株に導入した。前者導入株では鞭毛形成能が低下した。一方、後者導入株では顕著な鞭毛形成活性を示した。また、前者導入株をさらにmutagenizeした株ではNAPの発現量が極端に低下し、鞭毛形成能も元株よりさらに低下した。以上から、NAPがタンパク量依存的に、あるいはup-regulationに伴って鞭毛形成に関与する可能性が示唆された。以上は日本細胞生物学会第54回大会にて発表した。
    2.オオヒゲマワリ目におけるアクチンファミリーの存在様式とNAPの起源
    C. moewusii,とV. carteriについて、NAPホモログを同定した。前者については未同定だったアクチンホモログも同定した。分子系統解析の結果、NAPと2種のNAPホモログがアクチンとは起源を異にする、新グループを形成することが判った。V. carterのNAPはC. reinhardtiiのNAPと高い相同性を示し、両種がきわめて近縁であるとする説を支持した。以上は米国細胞生物学会第41回年会にて発表した。

    クラミドモナスには骨格筋アクチンと90%のアミノ酸相同性を持つアクチンと、それとは非常に異なる配列を持つNAP(アクチンとの相同性64%)の、二種のアクチン遺伝子が存在し、互いに排他的に発現している。一生物種内に保守的なアクチンと非保守的なNAPが共存している例はこれまでに報告がない。今年度は2種のアクチンの機能的差異を検討するために、分裂酵母発現系を用いた機能解析を行った。また、他生物種におけるNAPホモログの有無を検討した。
    1.アクチンとNAPの酵母発現モデルの作成・解析
    NAP、及びアクチン遺伝子を分裂酵母の発現用ベクター(pREP1、pREP81)に導入した。NAP-pREP1で形質転換した分裂酵母株は細胞質分裂が著しく阻害され、多核の長い細胞が多く現れた。2割程度の細胞では抗NAP抗体による染色で分列環に似たリング状構造が確認され、リング内に少量の内在性アクチンが共局在した。
    2.他生物種におけるNAPの存在の有無
    近縁種のボルボックスを用い、NAP遺伝子ホモログの有無をサザンブロット法により検討した。NAPのcDNAコード領域の中間部をプローブに用い、ハイブリダイゼーションを行ったところ、反応するバンドは全く検出されなかった。徐々にストリンジェンシーを下げると、ボルボックスのアクチン遺伝子に相当するバンドが薄く検出された。ボルボックス・アクチンとNAPのアミノ酸配列相同性は65%である。実験結果はこの生物に少なくともアクチン以上にNAPと相同性を持つようなNAPホモログは存在しないことを強く示唆した。これは両種のアクチンが98%の高い相同性を持つこととは対照的である。
